After administering low doses of the TLR4 ligand lipopolysaccharide (LPS) to mice, Shi et al. observed emigration of monocytes from the bone marrow to the blood. This response was dependent on TLR4 and myeloid differentiation primary response protein 88 (MYD88), an adaptor protein of the TLR signalling pathway. Moreover, studies in knockout mice indicated that CC-chemokine receptor 2 (CCR2) and its ligand CC-chemokine ligand 2 (CCL2; also known as MCP1) are also essential for the mobilization of bone marrow-residing monocytes.

In order to elucidate the mechanism of CCR2- and CCL2-mediated monocyte emigration, the authors generated CCR2- and CCL2-reporter mice. Thus, they could observe that CCR2+ monocytes are scattered in the bone marrow stroma during the steady state and move towards the vasculature after LPS administration. Furthermore, CCL2 expression was also localized to the blood vessels of the bone marrow.
