Treatment of solid tumours with chimeric antigen receptor (CAR)-T cell therapy has been challenging. One of the main difficulties faced is the lack of specific antigens. In addition, in those solid tumours that express specific antigens, this expression can be heterogeneous. Now, two papers published in Science Translational Medicine address these challenges by developing synthetic Notch (synNotch) CAR-T cells that target solid tumour-specific antigens and show efficacy in mouse models of glioblastoma, mesothelioma and ovarian cancer.
