Detection of cell-dependent cytotoxic antibody to cells infected with herpes simplex virus

Abstract

THE killing or damage of virus-infected cells by immune processes may play a significant role either in host defence or in the production of pathological lesions during virus infections1. These immune processes are believed to be directed at virus-induced antigens on the membranes of infected cells. On the basis of in vitro experiments, several mechanisms have been proposed as possibly operative in vivo. These include the cytolytic action of antibody and complement2,3, the interaction of antibody, complement and leukocytes4, and the destruction of infected cells by immune lymphocytes5–7. Another mechanism described in non-viral systems is the cytotoxicity demonstrated by the synergistic effect of antibody and nonimmune effector cells8–10. The following experiments demonstrate a similar process operative with viruses in which heat-inactivated immune serum augments the damage of herpes simplex virus-infected cells in vitro by human mononuclear cells.
