Abstract
DE DUVE et al. 1 have described the occurrence in rat liver of a new type of cytoplasmic particle, the lysosome, which was characterized by the following properties: (1) A distinct enzymic composition, containing a number of hydrolytic enzymes (acid phosphatase, β-glucuronidase, acid ribonuclease and acid deoxyribonuclease). (2) They can be isolated as a distinct intermediate fraction, between the mitochondria and microsomes, by differential centrifugation. (3) When isolated undamaged, the lysosomes show a low level of enzyme activity, but when the membrane is ruptured by freezing, and thawing, or by the addition of non-ionic detergents such as ‘Triton-X-100’, there is a considerable increase in the activity of the enzymes.
