The authors performed immunohistochemistry in post-mortem samples of human hippocampus at different stages of development to label putative neural progenitor cells (that is, expressing SOX1 or SOX2) undergoing proliferation (also expressing the proliferation marker KI67). Many KI67+SOX1+ cells and KI67+SOX2+ cells were observed in the dentate neuroepithelium (dNE) at 14 gestational weeks (14GW). At 22GW, these double-labelled cells were instead observed in the hilus or in the adjacent granule cell layer (GCL), albeit in lower numbers. KI67+SOX1+ cells and KI67+SOX2+ cells continued to drop in number in the first year of life, and were absent, or extremely rare, in the hippocampi of individuals aged 7 years or older. Moreover, these cells did not, at any developmental stage, form a discrete layer beneath the GCL, akin to the proliferative subgranular zone (SGZ) seen in rodents.
