Building actin networks on vesicles

RAB11A+ vesicles can move long distances towards the oocyte surface along actin filaments. To investigate the function of these vesicles, the authors expressed a dominant-negative variant of RAB11A in mouse oocytes. The resulting loss of RAB11A+ vesicles prevented asymmetric positioning of the spindle during meiotic division, which is required for the generation of a large, fertilizable egg. Asymmetric spindle positioning has been shown to require the actin network, which is normally highly dynamic but was static in the absence of RAB11A+ vesicles. As a result, the spindle was immobile and trapped in the static actin network. Indeed, when the actin network was artificially stabilized in normal mouse oocytes, asymmetric spindle positioning was also blocked.
