...NE assembly can, surprisingly, occur without the need for ATP or vesicle trafficking.

Fractionated cytosol and membranes from Xenopus laevis eggs can faithfully recapitulate NE assembly. Using this system, it was previously shown that NE formation can be blocked by the membrane-fusion inhibitor GTPγS or by the presence of non-hydrolysable ATP (ATPγS), which indicated that NE reassembly might be mediated by the active fusion of NE fragments. However, Anderson and Hetzer show that the application of GTPγS or ATPγS after the ER has reorganized does not affect NE formation (the ER becomes fragmented during the fractionation process). Therefore, NE assembly requires an intact ER rather than vesicle fusion or ATP consumption.
