In their comments on our paper1, Bjerknes and Cheng argue that cells staining positive for phosphorylated PTEN (P-PTEN), which retain 5-bromodeoxyuridine (BrdU) long-term and are located at positions 4 or 5 above Paneth cells, also express the marker chromogranin A (ChrA) and are therefore more likely to be enteroendocrine cells than intestinal stem cells (ISCs). They also question the reliability of the BrdU-retaining method and the related ISC position.
