Abstract
IN terms of a multiple-stage model of chemical carcinogenesis, tumour promoters may block differentiation of initiated (somatically mutated) cells and permit sufficient cell proliferation to overcome growth suppression1,2. Deacylation of cellular lipids, arachidonic acid metabolism, or both, are biochemical pathways that accompany the effects of tumour promoters and may even mediate cellular differentiation and carcinogenesis3. While investigating the effects of several putative anticarcinogenic compounds, we found and report here that retinoids, like the tumour-promoting phorbol diesters, stimulated deacylation of cellular lipids and prostaglandin production in dog kidney (MDCK) cells. Treatment of the cells with a phorbol diester and a trimethylmethoxyphenyl analogue of retinoic acid led to a synergistic stimulation of lipid deacylation and prostaglandin production.
