Transcription of a mammalian sequence under phage λ control

Abstract

RECOMBINANT DNA techniques have recently enabled bacterial plasmids carrying eukaryotic sequences syn-thesised in vitro from purified mRNAs to be constructed and cloned in Escherichia coli1–10. We show here that a rabbit globin sequence is not transcribed in E. coli when carried by the pCR1 plasmid. However, after transfer into phage λ, the globin sequence is transcribed under λ control. Thus, there is no basic hindrance to the transcription of a specific mammalian sequence in E. coli.
