Abstract
SYNAPTOSOMES prepared from the cerebral cortex by homogenization and density gradient centrifugation show a variety of metabolic properties characteristic of whole cells when incubated in suitable media1,2. Increasing the potassium concentration of the medium or electrical stimulation caused a preferential calcium dependent3 release of the physiologically active amino-acids glutamate, aspartate and GABA4. Acetylcholine synthesized by synaptosomes was also released by K+ and electrical stimulation5,6, and was also calcium dependent6. It has been shown7 that nerve-endings isolated from the median eminence of the hypothalamus contain corticotrophin-releasing factor (CRF) and antidiuretic hormone (ADH), and this study was undertaken to establish whether electrical and potassium stimulation would cause the release of neurosecretory substances and amino-acids from hypothalamic synaptosomes.
