Feedback Inhibition of Purine Biosynthesis de novo in Mouse Tissues in vivo

Abstract

THE inhibition of purine biosynthesis de novo in tumour cells by purines and purine analogues has been demonstrated by several workers1–3. The localization of the inhibition at the first reaction in this pathway, 5-phosphoribosyl-1-pyrophosphate-amidotransferase, was suggested by investigations with partially purified preparations of the enzyme from pigeon liver4–6 and bacteria7,8, and was supported by the results of investigations using intact tumour cells9. Preliminary work using crude rat liver extract agrees with these conclusions6.
