RNAi vector construction. RNAi feeding vectors used here were based on the pPD129.36 backbone (a kind gift from A. Fire, SUSM, Stanford, CA). The complete cDNAs of ufd-2 (using the forward primer 5′-ATGATTGAAGACGAGAAAGCAG-3′ and the reverse primer 5′-TTATTTCTTTGAATTTCTTTTCTG-3′) and chn-1 (using the forward primer 5′-ATGTCAAGCGGCGCCGAAC-3′ and the reverse primer 5′-TTAAACCATGCCTCCGGGTTC-3′) were amplified and subcloned into pPD129.36. RNAi constructs that are specific for individual depletion of cdc-48.1 or cdc-48.2, respectively, have been constructed as described previously1 (see also ref. 23 in the article).
