Error Frequency during in vitro Transcription of Poly U is increased with γ-Irradiated RNA Polymerase

Abstract

SINCE the original postulation of the “Central Dogma”1 it has been noted that changes in environment of the enzymes involved in replication, transcription and translation of the genetic message may induce error2–7. The most direct evidence is the increased mutation rate in DNA replication observed in gene 43 mutants of coliphage T4 which contain mutagenic DNA polymerase6. This increased error frequency has also been observed in vitro using mutagenic DNA polymerase7.
