An approach combining in situ tagmentation and transcription with MERFISH enables spatial profiling of the epigenome in tissues with single-cell resolution.

Tian Lu and Cheen Euong Ang in the laboratory of Xiaowei Zhuang at Harvard University sought to address this gap in methodology. “The epigenome, such as histone and DNA modifications, is critical for differential activation or repression of genes to generate different cell types. We were thus keen on expanding the repertoire of spatial omics tools and developing a spatially resolved single-cell epigenomic profiling method to help accelerate understanding of the spatial regulation of cell and tissue functions,” recalls Zhuang of their motivation to begin the work.
