Advanced systemic mastocytosis (AdvSM) is a rare haematological neoplasm in which mast cell accumulation leads to extensive damage in multiple organs, debilitating symptoms and median overall survival durations of <3.5 years. AdvSM is driven by the KIT D816V mutation in ~95% of patients. Now, data from two trials demonstrate the efficacy of the KIT D816V-selective inhibitor avapritinib in this disease.
