Synthesis of Poly(A) in vitro by Purified Virions of Vesicular Stomatitis Virus

Abstract

Poly(A) was first shown1 to be associated with cytoplasmic RNA in 1967. Since then it has been shown to be associated with many RNAs that function as messenger or as intranuclear mRNA precursors2–13. This includes the virion RNA of poliovirus7,8, eastern equine encephalitis virus7, Sindbis virus9,10 and RNA tumour viruses11–13. In addition, poly(A) is synthesized by cores of vaccinia virus in vitro2. The function of poly(A) tracts in mRNA has not yet been defined nor has the exact mechanism of synthesis and addition of poly (A) been elucidated, although the evidence favours post-transcriptional addition of poly(A) to mRNA4,7,14–16.
