Multiple Antigenic and Biological Determinants of Human FSH and its Subunits

Abstract

HERE we contrast the behaviour of human follicle stimulating hormone (hFSH) and its subunits (hFSHα, hFSHβ) in four different in vitro systems. The markedly non-uniform patterns observed with respect to hFSH and its subunits in three different immunoassay systems and in a receptor assay system lead us to conclude that multiple antigenic and biologic determinants exist on the hFSH molecule. In all studies we used hFSHα (LER 1661 (8–10)), hFSHβ (LER 1679-82) and hFSH (LER 1366) except in the receptor assay system where the second International Reference Preparation (IRP) of human menopausal gonodotrophins (2nd IRP-HMG) was used instead of LER 1366. The bioactivity of 1 ng LER 1366 is equal to 3 mIU 2nd IRP-HMG so that in terms of weight of hFSH, 1 mIU 2nd IRP-HMG is equivalent to well under 1 ng of hFSH.
