Effects of Combining Ultraviolet Repair and Recombination Mutations in Haemophilus influenzae

Abstract

IN order to determine effects of double mutations at loci affecting recombination and repair of damage caused by ultraviolet radiation in Haemophilus influenzae, we have attempted to combine mutations from strains isolated previously in this laboratory1,2. Our most significant result is that we were unable to isolate a strain with a mutation in both the rec1 gene and the uvr1 gene. The rec1 gene makes the cell recombination-defective and also ultravioletrsensitive due to a defect in post-replication repair3. The uvr1 mutant is ultraviolet-sensitive because of a defect in the specific endonuclease operating during the excision of ultraviolet-induced pyrimidine dimers4. We conclude that both gene products are used for normal maintenance of DNA in H. influenzae, and that this combination of mutations is lethal.
