Inhibition of E. coli DNA Polymerase II by Ara-CTP

Abstract

THE potent antileukaemic agent, 1-β-D-arabinofuranosylcytosine (ara-C), specifically inhibits DNA synthesis in bacterial and animal cells1,2. Although the exact mechanism of inhibition has been in doubt, it seems likely that it occurs at the DNA polymerization reaction itself2–5. We describe here the effects of ara-CTP, which is the most prominent form of ara-C in the cell, on in vitro replicative systems6–11 of E. coli and on isolated DNA polymerases II and III12–15.
