Molecular glue-like STING activator

Stimulator of interferon genes (STING) is a dimeric transmembrane protein in the endoplasmic reticulum that plays an essential role in the innate immune system, inducing a variety of pro-inflammatory cytokines and chemokines, including type I interferons (IFNs). STING is normally activated by the binding of cyclic di-nucleotides (CDNs) to the cytosolic ligand binding domain (LBD), which induces a conformational change in STING that enables the formation of a high-order oligomer. CDN analogues and compounds binding to the LBD of STING are being explored in cancer clinical trials.
