C17orf80 joins nucleoids

Mitochondrial DNA (mtDNA) replication is important to sustain mtDNA levels and support mitochondrial functions. How mtDNA replication is regulated is still poorly understood. Wu et al. find that the previously functionally uncharacterized protein C17orf80 promotes mtDNA duplication.

Compact mtDNA molecules and associated proteins form nucleoids in mitochondria, and perturbations in nucleoid components or mutations in mtDNA are linked to disease. Wu et al. used mass spectrometry to profile proteins interacting with mtDNA in isolated mitochondria from human cells. Electron microscopy, biochemical studies and chromatin immunoprecipitation with quantitative real-time PCR showed that C17orf80 localizes to mitochondrial nucleoids and binds to mtDNA without sequence specificity. C17orf80-depleted cells had lower levels of mtDNA, signs of impaired mtDNA replication, and perturbed cell proliferation and mitochondrial metabolism.
