Activation of Phosphofructokinase by Stimulants of Cell Multiplication

Abstract

THE growth of chick embryo fibroblasts (CEF) in culture is markedly slowed by reducing the pH of the medium and removing serum1, 2. DNA synthesis is gradually reduced in the culture as a whole due to an increase in length of the G1 period and consequent decrease in the fraction of cells making DNA at any time. This fraction increases a few hours after initial pH and serum concentrations have been restored1. Sugar transport is similarly but more quickly affected by alterations to the medium1. These events simulate the effects of cell population density on the growth of cells3. Manipulation of the cells by varying pH and serum concentration therefore offers a well defined way of studying the physiological basis for growth regulation.
