The HIV-1 accessory protein Nef plays an essential role in HIV-1 pathogenesis, enhancing viral infectivity and promoting immune escape, making it an attractive antiretroviral drug target. Although several studies have reported drug discovery efforts targeting HIV-1 Nef, these have been complicated by the fact that Nef lacks intrinsic enzymatic activity and an active site.
