Stalled molecular motor inhibits tumour growth

Aggressive cancers have high frequencies of chromosomal instability (CIN), arising from defects in the mitotic spindle and in chromosome segregation during cell division. Therapeutic targeting of CIN is an emerging approach to exploit cancer-specific vulnerabilities. In Nature Cancer, Payton et al. now report potent small-molecule inhibitors of the mitotic kinesin KIF18A and demonstrate their activity in mouse models of cancers with CIN.
