Abstract
MATURING oocytes of the frog Rana pipiens arrest in prophase of the first meiotic division, when they can be isolated and induced to complete meiotic maturation in vitro by exposure to steroid hormone. A study of the meiotic maturation divisions of the frog oocyte therefore encompasses problems of steroid action and the control of cell division. An unusual feature of the interaction of steroids with the oocyte is the independence of the nucleus indicated by the failure of actinomycin D or enucleation to affect the induction of maturation by steroids1. Furthermore, the interaction is nonspecific, in that several different steroids are capable of inducing maturation in vitro2. Both cyclic AMP and theophylline inhibit progresterone induced maturation3. This report presents evidence that exposure of prophase-arrested oocytes to progesterone lowers intracellular cyclic AMP and cyclic GMP levels, that theophylline inhibits one component of this response, and that periodic cyclic nucleotide fluctuations occur during the subsequent maturation divisions.
