The adjuvant active fraction of delipidated mycobacteria

Abstract

FREUND's complete adjuvant (FCA) has been used to influence both cell-mediated and humoral immune responses. Mycobacteria are the essential constituents of FCA and different active fractions have been extracted from mycobacteria of various strains1–6. The purpose of the work reported here was to try to answer the following question : are cell-mediated and humoral immune responses related to the stimulatory effect of different bacterial components or to the adjuvant activity of a same bacterial fraction? Data presented here suggest that the stimulation of these two kinds of immunological response is related to the adjuvant activity of the same bacterial fraction, which has been identified as the lipoglycopeptide fraction extracted from mycobacterial delipidated cells (LG). Furthermore, the study of the relationship between adjuvant activity and chemical structure showed that LG is comprised of two distinct moieties: a glycopeptide moiety with cellular activity and a lipid moiety stimulating humoral immune response.
