Platelet 5-HT uptake and release stopped rapidly by formaldehyde

Abstract

UPTAKE of 5-hydroxytryptamine (5-HT) and the thrombin-induced release reaction in platelets are relatively rapid processes at 37 °C, with a linear component of approximately 1 min for uptake1 and an estimated half life of 6 s for the release reaction2,3. An agent which could essentially stop these processes instantaneously and prevent any further 5-HT exchange across the plasma membrane would therefore be a valuable tool for precise studies of the kinetics of uptake and release. We describe here the treatment of platelets with formaldehyde, a technique which seems to provide such a tool.
