Abstract
THE final step in assembly of IgA and IgM is the polymerisation of intracellular monomeric (H2L2) subunits. This event takes place just before, or at the time of secretion1–7, and involves the incorporation of J chain8,9 into IgM10 and polymeric, but not monomeric IgA10–11. An unanswered question is whether J chain travels through the membranous elements of the cell, as do the other immunoglobulin subunits12. As a prelude to answering this, we have demonstrated intracellular J chain in polymer-secreting mouse myeloma cells. In addition, the postulation of an intraclonal switch from IgM to IgG in B-lymphocytes13 prompted us to look for, and find, J chain in cells which secrete IgG.
