BARCODE-ALL: accelerated and cost-effective genetic risk stratification in acute leukemia using spectrally addressable liquid bead microarrays

A single tube assay capable of automating and expediting the detection of hybrid mRNA transcripts in pediatric acute lymphoblastic leukemia is described. The assay combines multiplex RT-PCR for simultaneous amplification of all relevant transcripts and fluid bead microarrays for simultaneous transcript identification.

Cells were isolated from peripheral blood or bone marrow aspirates by dextran-assisted red blood cell sedimentation. Archival samples were washed in 1 × PBS before RNA preparation.
