Abstract
THE isolation and purification of dopamine β-hydroxylase1,2 and the elucidation of the role in the biosynthesis of nor-adrenaline has been described already. The susceptibility of the enzyme to chemical inhibition has stimulated widespread interest in the use of enzyme inhibition in vivo as a pharmacological tool or an antihypertensive drug. Disulphiram, through its reduced metabolite diethyldithiocarbamate (DDC), is an effective inhibitor of dopamine β-hydroxylase3,4. No inhibitors of dopamine β-hydroxylase had been used successfully in the treatment of hypertension in man until 5-butylpicolinic acid was tried.
