Opportunities and challenges with hyperpolarized bioresponsive probes for functional imaging using magnetic resonance

Bioresponsive hyperpolarized probes contain magnetic resonance signals that can be many orders of magnitude larger than those of common, thermally polarized probes. This Perspective discusses how bioresponsive hyperpolarized probes can be directly linked to biological events to give functional information, enabling the mapping of physiological processes and diseases in real time using magnetic resonance.

A versatile bioelectronic interface programmed for hormone sensing

Ultrasensitive, real-time profiling of bio-analytes is a prerequisite for precision medicine. Here, the authors present a versatile bio-electronic interface (VIBE) to sense signaling cascade-guided receptor-ligand interactions and show that it can detect hormone levels in blood samples and differentiate individual metabolic conditions.

A smart pathogen detector engineered from intracellular hydrogelation of DNA-decorated macrophages

Macrophages have the inherent capacity to recognise pathogenic microorganisms, highlighting their potential to be used in biosensing applications. A highly sensitive and robust one-pot assay for the detection of microRNAs

We developed exponentially amplified rolling circle amplification with CRISPR–Cas12a as a one-pot, isothermal assay for microRNA detection. This method has single-digit femtomolar sensitivity and single-nucleotide specificity, and can be deployed for point-of-care testing. The assay has been adapted for the microRNA profiling of extracellular vesicles, which is used in the diagnosis of pancreatic cancer.

A brilliant monomeric red fluorescent protein combining high brightness and fast maturation

We evolved the brilliant monomeric red fluorescent protein mScarlet3 using a multiparameter screening approach. Owing to a newly engineered hydrophobic patch inside its β-barrel structure, mScarlet3 combines a high quantum yield and high fluorescence lifetime with fast and complete maturation. Consequently, mScarlet3 performs well as a fusion tag in live-cell imaging.
