RHO signalling is a cell signalling pathway by which extracellular or intracellular stimuli promote activation of the small GTPase RHO. Cell adhesion, mechanical stress and membrane receptor signalling activate RHO, which signals through effectors to promote changes in cell adhesion and contractility, gene expression, cell division and proliferation, and other effects.

Deficiency of gluconeogenic enzyme PCK1 promotes metabolic-associated fatty liver disease through PI3K/AKT/PDGF axis activation in male mice

Phosphoenolpyruvate carboxykinase 1 (Pck1) is an enzyme involved in glucose production that also regulates lipogenesis and has been linked to liver steatosis. Here the authors report that deficiency of Pck1 in the liver leads to nonalcoholic fatty liver disease via activation of the RhoA/PI3K/AKT pathway in a study with male mice.

Interaction kinetics between p115-RhoGEF and Gα₁₃ are determined by unique molecular interactions affecting agonist sensitivity

The three RH-RhoGEFs differ in their sensitivity of activation by G proteins. For p115-RhoGEF, a specific molecular interaction with Gα₁₃ is responsible for faster kinetics and lower sensitivity.

SH3 domain regulation of RhoGAP activity: Crosstalk between p120RasGAP and DLC1 RhoGAP

Inhibition of DLC1 RhoGAP by p120RasGAP allows for crosstalk between the Rho and Ras pathways. Here, Chau et al. present the co-crystal structure of DLC1 RhoGAP domain and p120RasGAP SH3 domain and probe the interaction with biochemical studies.

EGFR ligands dictate tumour suppression

EGFR is an oncogene that is frequently amplified in glioblastoma. A new study suggests a tumour-suppressive role of EGFR in EGFR-amplified glioblastoma regulated by ligand abundance. Increased EGFR ligand in EGFR-amplified glioblastoma suppresses invasion by upregulating BIN3 and inhibiting activation of Rho GTPases.

Endothelial metabolism going single

Endothelial cell migration is indispensable for (tumour) angiogenesis and is fuelled by glycolysis. The metabolic underpinnings have been only partially unravelled to date, partly because of the lack of appropriate tools to analyse metabolic flux at the single-cell level. In this issue, Wu et al. introduce a novel imaging assay to capture glycolytic flux during motion in single endothelial cells. With this new tool, the authors uncovered a glycolysis-driven cytoskeletal remodelling apparatus that propels endothelial cell motility.
