Raman spectroscopy

Raman spectroscopy is an optical technique that detects intrinsic vibrational, rotational and other low-frequency modes in molecules upon inelastic scattering of monochromatic light. Because different chemical functional groups scatter light at different frequencies, Raman spectroscopy can be used as a tool for chemical structure analysis, chemical fingerprinting and chemical imaging.

    Samuel, Miyaoka et al. investigate the changes in the molecular composition of lipid droplets inside HuH7 cells and its relation to the physiological state of the cell, using Raman spectroscopy and multivariate curve resolution. This study underscores the importance of separation of spectral components in semi-quantitative molecular profiling of living cells.

    Most current cell sorting methods are based on fluorescence detection with no imaging capability. Here the authors generate and use Raman image-activated cell sorting with a throughput of around 100 events per second, providing molecular images with no need for labeling.

    The authors develop a first-principles workflow for calculating Raman spectra of 733 monolayers from the computational 2D materials database. After benchmarking results against experimental data for 15 monolayers, an automatic procedure for identifying a material from its Raman spectrum is proposed.
