Parathyroid diseases

Parathyroid diseases are disorders of parathyroid gland function, resulting in abnormally high (primary hyperparathyroidism) or low (primary hypoparathyroidism) levels of calcium and parathyroid hormone. Secondary hyperparathyroidism is characterized by normal calcium but high parathyroid hormone levels, whereas pseudohypoparathyroidism is associated with low calcium and high parathyroid hormone levels.

An optogenetic approach for regulating human parathyroid hormone secretion

Parathyroid hormone (PTH) plays a role in maintaining calcium and phosphorus homeostasis, and in secondary hyperparathyroidism excess PTH secretion contributes to bone loss. Here the authors report an optogenetic approach to inhibit PTH secretion in human hyperplastic parathyroid cells, and prevented hyperplastic parathyroid tissue-induced bone loss in mice.

Asymmetric activation of the calcium-sensing receptor homodimer

Cryo-EM structures of human calcium-sensing receptor reveal intrinsic asymmetry in the receptor homodimer upon activation that is stabilized by calcimimetic drugs adopting distinct poses in the two protomers, priming one protomer for G-protein coupling.

PTH hypersecretion triggered by a GABA_B1 and Ca²⁺-sensing receptor heterocomplex in hyperparathyroidism

Parathyroid glands regulate calcium homeostasis by secreting parathyroid hormone. Chang et al. demonstrate that in parathyroid glands GABA receptor GABAB1R makes a heterocomplex with the calcium-sensing receptor and regulates the secretion of parathyroid hormone.
