Crystal structures reveal catalytic and regulatory mechanisms of the dual-specificity ubiquitin/FAT10 E1 enzyme Uba6

Uba6 is an E1 enzyme that regulates numerous cellular processes by activating ubiquitin and FAT10 pathways. Here, the authors present crystal structures that illuminate Uba6 catalytic mechanisms and reveal inositol hexakisphosphate as a cofactor that modulates Uba6 activity.

Engineered Cas13 variants with minimal collateral RNA targeting

The application of CRISPR–Cas13 for targeted RNA degradation is hindered by the existence of collateral effects induced by Cas13. We screened hundreds of engineered Cas13 variants using a convenient and sensitive reporter system and obtained several variants with markedly reduced collateral activity and efficient on-target activity, suitable for in vivo applications.

Landscape of somatic mutations in aging human heart muscle cells

Using single-cell whole-genome sequencing, we identified and characterized the landscape of somatic single-nucleotide variants (sSNVs) in single cardiomyocytes from individuals across the human lifespan. Aged cardiomyocytes were found to have a higher burden of sSNVs and show mutational signatures that suggest failed repair of oxidative DNA damage.

Filling in the blanks: how the C-strand catches up to the G-strand at replicating telomeres using CST

CST is both an ssDNA-binding complex and a DNA Pol-α/primase cofactor that coordinates the switch from G-strand elongation to C-strand fill-in during telomere maintenance.
