Chromosome instability describes the increased probability of acquiring chromosomal aberrations due to defects in processes such as DNA repair, replication or chromosome segregation.

    Ranhja et al. find that budding yeast Dna2 is modified by sumoylation, and that sumoylation reduces the nuclease, but not helicase activity of Dna2 in vitro. In cells, expression of a Dna2 version that cannot be sumoylated leads to impaired DNA end resection and cell division.

    Chordomas are rare bone tumors with limited therapeutic options. Here, the authors identify molecular alterations associated with defective homologous recombination DNA repair in advanced chordomas and report prolonged response in a patient treated with a PARP inhibitor, which later acquired resistance due to a newly gained PARP1 mutation.
