Structural basis for SHOC2 modulation of RAS signalling

Cryo-electron microscopy structure, molecular dynamics and biochemical analyses of the SHOC2–PP1C–MRAS complex demonstrate the dependence of the complex formation on RAS–GTP and identify the determinants of RAS isoform preference for SHOC2–PP1C and specificity of the complex for RAF dephosphorylation.

Targeting drug-resistant mutations in ALK

Therapy resistance limits the clinical success of tyrosine kinase inhibitors (TKIs) in ALK-positive non-small cell lung cancer. A study now proposes a framework to identify compound resistance mutations to the lorlatinib TKI and provides structure-based drug design approaches to overcome resistance mediated by ALK(G1202R) or ALK(I1171N/S/T).

Efficacy of antiangiogenic medication to treat arteriovenous malformations

In a prospective experimental observational study of 18 individuals with severe, recurrent extracranial arteriovenous malformations, thalidomide was effective in reducing pain, healing ulceration, stopping bleeding and resolving cardiac failure.
