Barrett oesophagus

Barrett oesophagus is a condition in which the squamous epithelium of the distal oesophagus is replaced by columnar epithelium via metaplasia, in this case intestinal metaplasia (goblet cells present). Barrett oesophagus is a precursor to oesophageal adenocarcinoma. The principal cause of Barrett oesophagus is chronic acid reflux.

Somatic whole genome dynamics of precancer in Barrett’s esophagus reveals features associated with disease progression

Barrett’s esophagus is a pre-malignant condition that can progress to esophageal cancer. Here, the authors carry out whole genome sequencing of samples from patients who did or did not progress to cancer and find that mutations in many genes occur regardless of progression status, but also find features associated with progressive disease.

Multi-omic cross-sectional cohort study of pre-malignant Barrett’s esophagus reveals early structural variation and retrotransposon activity

The alterations associated with progression from Barrett’s esophagus (BE) to esophageal adenocarcinoma are not fully characterised. Here, the authors perform a multi-omics analysis of a longitudinally-sampled BE patient cohort, identifying the impact of structural variants, including mobile elements, and the timing of molecular events during progression.

Evolutionary dynamics in Barrett oesophagus: implications for surveillance, risk stratification and therapy

Cancer development is a dynamic evolutionary process. This Review explores the mechanisms underlying clonal expansion and contraction as well as the evolutionary dynamics in Barrett oesophagus and its progression to dysplasia and oesophageal adenocarcinoma. The implications for patient management (including surveillance, risk stratification and therapy) are also explored.

Molecular epigenomic subtypes of Barrett neoplasia

Genomic alterations are well-described in Barrett oesophagus and its sequela, oesophageal adenocarcinoma, but epigenomic events have not been so widely investigated. In a new study, Jammula et al. analysed the DNA methylation profiles of Barrett oesophagus, oesophageal adenocarcinoma and control tissues. Four molecular subtypes were identified, revealing four distinct epigenomic classes and potential therapeutic targets.

Dr. Kate Miroshnikova is a bioengineer by training and is currently a postdoctoral EMBO/HFSP fellow at the Max Planck Institute for Biology of Ageing and at the Helsinki Institute of Life Science. She is interested in understanding biomechanical regulation of stem cell fate decisions in health and disease. Kate’s long term scientific interest is to understand how cells and tissues sense, integrate, and adapt their transcriptomes and proteomes to the highly dynamic mechanical environments without compromising structural and genomic integrity.
