A postdoctoral associate research position is available immediately for a highly motivated scientist to join the Synthetic Biology Center at MIT in a sponsored research project at the intersection of cell fate reprograming, mammalian synthetic biology, and feedback control. This involves, working efficiently and cooperatively with a team of researchers and the lab manager to participate in a sponsored research program in synthetic biology. Responsibilities include verbal and written communication of results to team members and presentation to the scientific community through talks and publications.

S/he will join the labs of Jim Collins (Biological Engineering, MIT, http://collinslab.mit.edu/) and Ron Weiss (Biological Engineering, MIT, http://groups.csail.mit.edu/synbio/), and a team including Domitilla Del Vecchio (Mechanical Engineering, MIT, https://scripts.mit.edu/~ddv/index.php ) and Thorsten Schlaeger (Human Embryonic Stem Cell Core Facility, Harvard Medical School).

In this project, we seek to develop a new and general approach to cell fate reprogramming based on the design of synthetic genetic feedback control circuits in mammalian cells. These circuits will have unprecedented ability to accurately control endogenous transcription factors’ concentration in the face of noise and endogenous gene regulatory network dynamics. As a first application, we will consider induced pluripotent stem cell (iPSC) reprogramming with an aim of improving currently low efficiency, kinetics of reprogramming, and quality of produced cells. The project will then consider other cell fate conversion and directed differentiation tasks. Overall, the project will make substantial contributions to stem cell biology while at the same time developing a general-purpose technology for cell fate conversion tasks.

The research will focus on the use of the genetic feedback controllers in cell fate reprogramming tasks. Responsibilities include virus production and titration, infection, lineage tracing, next-generation sequencing, bioinformatics analysis, flow cytometry, live imaging, antibody staining, qRT-PCR, mammalian and stem cell culture, iPSC reprogramming of human somatic cells, maintenance of partially and fully reprogrammed human iPSCs, in vitro differentiation assays, closely cooperating with a student with a modeling/quantitative background and with a postdoctoral associate with a synthetic biology background; and presenting to the scientific community through talks and publications.

Requirements:
• Ph.D. in biological sciences, engineering, or related field.
• Experience in mammalian cell culture, stem cells, next-generation sequencing.
• Biological experience is required in cell biology.
• Expertise in developmental biology and skills in flow cytometry.
• Highly motivated individual who can prioritize and perform multiple tasks in a dynamic, multidisciplinary, team-oriented setting.
• Strong analytical, problem-solving, documentation, organizational, and interpersonal skills.
• Ability to execute detailed technical protocols meticulously.
• A minimum one-year commitment is required.
