About the role
We wish to recruit a Postdoctoral Research Associate (PDRA) to the group of Dr Pietro Roversi, to take a central role in a research project to investigate rescue of secretion of rare disease-associated glycoprotein mutants in WT and UGGT1 knock-out mammalian cells.
The research is funded by a Wellcome Trust Seed Award in Science and will constitute the first ever test of the hypothesis that the secretion of slightly misfolded, and yet active human glycoproteins, can be rescued in mammalian cells by the deletion of the eukaryotic glycoprotein folding checkpoint, the enzyme UGGT1. The study will involve cloning of fluorescent fusion glycoprotein constructs, transfection of those constructs into mammalian cells and confocal microscopy to follow the fluorescent fusion glycoproteins as they traverse the secretory pathway of the cells. The secretome of the WT and UGGT1 knock-out mammalian cells will also be characterised by mass-spec proteomics. You will take a prominent role in designing the experiments, along with shaping the direction of the programme and will develop your own research ideas in line with the aims of the research programme. You will be encouraged to develop your own career, engaging in preparation towards future stages of your professional life. You will write up your research findings for dissemination amongst the research team and the broader international community and make presentations at scientific meetings in the UK and overseas.
The successful applicant should hold a PhD, have a strong background in molecular biology, mammalian cell culture and fluorescence microscopy and have a keen interest in experimental design.
