A PhD student position is available in the research group of Prof. Berend Isermann (previously at Otto-von-Guericke University Magdeburg) from the Institute of Laboratory Medicine at Leipzig University. The project aims at studying the role of coagulation proteases dependent signal transduction for renal disease. The main focus of the laboratory is the role of coagulation proteases in vascular complications. Our particular focus is diabetic nephropathy (Isermann, Nature Medicine, 2007; Wang, Isermann, Blood, 2011; Madhusudhan, Isermann, Blood, 2012; Bock, Isermann, PNAS, 2013; Shahazd, Isermann, KidInt, 2014; Madhusudhan, Isermann, Nature Comm., 2015; Dong, Isermann, JASN, 2015; Shahzad, Isermann, JASN, 2016; Kohli, Isermann, Blood 2016; Madhusudhan, Isermann, Blood, 2017). Within the current project, novel aspects of cellular metabolism, epigenetic regulation, cell-plasticity and the relevance of this for subcellular localisation of receptor complexes and organelle function (mitochondria, ER) will be analysed.

The project involves:

  • Applying various methods of ex vivo analysis, including co-immunoprecipitation, proteomics, proximity ligation assay, mitochondrial metabolism, ChIP-on-chip assay and single-cell sequencing.
  • Animal experiments with transgenic mice as well as therapeutic interventions, surgeries.
  • Investigation of epigenetic changes and mechanisms of metabolic reprogramming in context to the coagulation system (in vitro and in vivo)
  • Investigation of mouse models with targeted mutations in the coagulation system, as well as
  • Microscopic analysis techniques such as immunohistochemistry, immunofluorescence and electron microscopy

Requirements:

  • Successful completion of master’s degree in life sciences (biology, biochemistry, biotechnology).
  • Relevant research experience (preferably international) and fluent English communication skills.
  • Comprehensive detailed knowledge in molecular biology, cell culture, mRNA, protein and DNA analysis, epigenetic modifications, as well as in vitro techniques in cell culture, cell migration, cell proliferation, co-culture models and cell death are desirable.
  • Knowledge in the field of coagulation dependent signal transduction and diabetic nephropathy is preferable.
  • Experience in the independent manipulation of mice (injections, organ removal) is required.
  • Experience in the application of cloning techniques including Crispr / Cas9 is preferable.
