Position Description

The Moses Laboratory, established in 2020 at Cold Spring Harbor Laboratory (CSHL), is the first chemistry focused laboratory at CSHL, led by the renowned Prof. John Moses. Known for advancements in click chemistry reaction development and application, the Moses Laboratory employs cutting-edge click reactions such as Sulfur Fluoride Exchange (SuFEx) and Phosphorus Fluoride Exchange (PFEx) to pioneer novel anticancer therapies. The Moses Laboratory’s primary objective is to identify functional molecules efficiently, working in collaboration with a network of biologists at CSHL to evaluate hit compound potencies and thoroughly probe underlying mechanisms of action. These studies often involve the synthesis of bespoke chemical probes and conducting protein pulldown experiments to identify therapeutic targets. By integrating chemistry and biology, the Moses Laboratory aims to accelerate the development of novel therapies for aggressive cancers, including pancreatic ductal adenocarcinoma and triple-negative breast cancer.

We are seeking a postdoctoral researcher with a background in biology to complement and support the ongoing chemistry work in our laboratory. The ideal candidate will possess significant expertise in protein target identification and purification, coupled with proficiency in executing cell-based assays such as cytotoxicity evaluations in both 2D and 3D cancer models. Experience in generating knockout cell lines to validate suspected mechanisms of action is highly desirable. The successful candidate will work collaboratively with other distinguished members of the CSHL community to advance our cutting-edge research programs.

Position Requirements

Education: Candidates must hold a PhD degree.
Skills: • Experience evaluating small molecules for cytotoxicity in human cell culture (e.g., determining IC50 and AUC values) • Experience conducting biophysical assays to probe underlying mechanisms of action (e.g., receptor engagement and enzyme inhibition assays). • Proficiency in purifying proteins, conducting protein target identification, and associated pulldown experiments. • Ability to generate knockout cells and auxin-inducible cell lines. • Proficiency with qPCR and western blot techniques. • Experience with analyzing data using relevant software (e.g., Prism).
Experience: • Live cell imaging. • Flow cytometry. • Generation of recombinant proteins. • Ability to perform Tier 1 ADME screens/use relevant kits – (Metabolism kits, Caco-2, CYP inhibition/induction). • Xenograft mice models.
