Proteomic identification of P. aeruginosa proteins that interact with laminin. Two-dimensional (2D) separation of outer membrane proteins from P. aeruginosa PAO1 by 2D-SDS-PAGE (a,b), and electrotransferred onto membrane blots that were subsequently incubated with laminin-111, rabbit anti-laminin pAbs and HRP conjugated swine anti-rabbit pAbs (c,d). Panels e and f comprise the background control of primary and secondary pAbs. Laminin-binding protein spots were excised from gels and identified by sequencing using MALDI-TOF, of which four were confirmed laminin-binding outer membrane proteins, i.e. EstA, OprD, OprG and PA3923. Numbers and arrows point toward each identified protein. The numbering and asterixis correspond to Table S3 in supplementary data. Denoted numbers to the right of the protein ladder in each picture indicate size in kilodalton. pI and numbers denoted at the top of each picture indicate the isoelectric point. All images are of full-length gels or immunoblots. Brightness and contrast have been adjusted over entire images to harmonize background levels.
