Key signaling events that lead to fetal membrane rupture following activation of stretch-activated pro-inflammatory pathways. The application of cyclic tensile strain (CTS) induced AKT and Cx43 protein expression leading to enhanced levels of PGE2 release and MMP activity. In contrast, CTS enhanced GAG synthesis which causes swelling and disruption of the extracellular matrix components via inhibition of collagen and elastin levels. An improved understanding of these mechanisms could lead to the further development of therapeutics for preventing PPROM.
