The regulation of HIF-1α. (A) In normoxic condition, Prolyl hydroxylase (PHD), in the presence of Fe2+ and oxygen molecule, hydroxylates HIF-1α and triggers its further ubiquitination and proteolysis. (B) In hypoxic condition or with the help of hypoxic mimicking agents such as FG-4592, the hydroxylation and degradation of HIF-1α are blocked. Thus HIF-1α can dimerize with HIF-1β, enter the nucleus to transcriptionally regulate the expression of its target genes such as VEGF, PDGF and vWF, ultimately regulating a wide range of pathophysiologic processes including angiogenesis.
