Propranolol and 2DG synergize in inducing arrest of proliferation and cell death of cancer cells. PC3 cells were treated with increasing concentrations of 2DG (1, 2 and 10 mM), propranolol (P) (100 µM) or both. (a) The proliferation of cells was measured by DNA content quantification as indicated in “Materials and Methods”. 2DG and P synergize to arrest PC3 cells. (b) Phase contrast microscopy photographs show the status of PC3 cells after 48 h of treatments (scale bar = 250 µm). (c,d) Cell death was measured by FACS analysis after labeling cells with FITC-annexin-V and propidium iodide. 10 000 events were collected for each experiment. (c) PC3, LnCaP and PNT1A were treated with 2DG (10 mM), propranolol (P) (100 µM) or both for 48 h. (d) MDAMB231 and 4T1 cells were treated with 2DG (10 mM and 2 mM respectively), propranolol (P) (100 µM and 50 µM respectively) or both for 48 h. Propranolol and 2DG synergize in inducing cancer cell death.
