MIDN localizes primarily to the nucleus and intracellular membranes in PC12 cells. (a) PC12 cells were incubated in the presence or absence of 100 ng/ml NGF for one day before being stained with anti-MIDN antibody. Localization of endogenous MIDN was observed using a confocal laser scanning microscope. MIDN and nuclei were colored in red and blue, respectively in the merge image. Scale bar: 10 µm (b) Following the NGF incubation described above, cells were homogenized and fractionated into nuclear, cytosolic, and crude membrane fractions as described in Methods. The MIDN in these fractions was then examined by Western blotting.
