Reporting in Nature, Xiao et al. describe the structure of the NLPR3–NEK7–ASC complex, paving the way to understanding inflammasome assembly and autoinflammatory disease.

The authors recombinantly expressed and purified NLRP3, NEK7 and ASC pyrin domain for in vitro reconstitution of the complex trapped in the active state with an ATP analog. Using cryo-EM, they obtained a 3D reconstruction of the complex.
