Single-nuclei isoform RNA sequencing enables cell-type specific isoform analysis in frozen brain samples.

RNA splicing, transcription start site and poly(A) site choice have fundamental roles in mRNA maturation and gene expression. Long-read sequencing has been previously used to study RNA isoforms in single cells. Yet it is not easy to isolate individual cells from frozen tissues. “While the words ‘frozen tissues’ may not be fundamentally inspiring, they englobe 99% of all human brain tissue samples,” says Hagen Tilgner from Weill Cornell Medicine in New York.
